The Barbarian Combat Masteries are passive skills that increase various attributes of the Barbarian. For example if you wanted to specialize in Swords, you would use Sword Mastery and then if you wanted to have more defense rating % you would specialize in Iron Skin.
More than one mastery can be used at one time. You can have points in both Axe Mastery and Mace Mastery. These skills have their origins in the Barbarian clans.
